摘要
目的探讨参芪扶正注射液对心肌挫伤后家兔心肌细胞凋亡及其Bcl-2和Bax蛋白表达的影响。方法健康家兔50只,简单随机化摸球法分为4组,参考组(Reference,n=20)、对照组(Control,n=10)、参芪扶正注射液治疗组(Sen,n=10)和心肌挫伤组(MC,n=10)。利用BIM-Ⅱ型生物撞击机制备心肌挫伤模型,应用ELISA法测定心肌肌钙蛋白I(cTnI),应用TUNEL法并结合流式细胞术观察心肌细胞凋亡发生的情况,应用免疫组化SABC法观察Bcl-2和Bax蛋白的表达。对照组除不撞击胸部外,余处理均同MC组。结果MC组与Sen组在伤后4h血清cTnI含量均高于各自伤前及对照组相应时相点(P<0.05);对照组未见TUNEL阳性细胞,MC组和Sen组心肌细胞凋亡指数(AI)分别为(15.6860±0.6887)%和(5.5790±0.1917)%(P<0.05),凋亡率分别为(6.9320±0.6380)%和(3.0210±0.2540)%(P<0.05);与对照组比较,MC组的Bcl-2蛋白和Bax蛋白表达光密度值均明显增高(P<0.05);而Sen组的Bcl-2蛋白表达光密度值高于MC组(P<0.05),Bax蛋白表达光密度值低于MC组(P<0.05)。结论参芪扶正注射液能有效的减少或抑制钝性心肌挫伤家兔心肌细胞的凋亡,对钝性心肌挫伤具有一定的保护作用,其抑制凋亡可能是通过上调Bcl-2蛋白表达和下调Bax蛋白表达来实现的。
Objective To study the effects of Senqi Fuzheng injection on the apoptosis of myocardial cells and the expressions of Bcl-2 and Bax in experimental rabbits with myocardial contusion (MC). Methods Totally 50 healthy rabbits were randomized into 4 groups: reference group (n = 20), control group (n = 10), Sen group (n : 10) and MC group (n : 10). MC model was inflicted with aid of BIM-Ⅱ biomedical impact-machine. The apoptosis of myocardial cells was analyzed by TUNEL. The expressions of Bcl-2 and Bax were detected by using immunohistochemistry with SABC method. The rate of apoptosis in myocardial cells was quantified with flow cytometry. Control group was treated similar to the MC group except the impact. Results The levels of serum cTnI were significantly increased 4 h after impact in MC and Sen group (P 〈 0.05 ). Myocardial cell apoptosis was only detected in MC and Sen group, with the apoptosis index (AI) of (15. 686 0 ±0. 688 7 ) % and ( 5. 579 0 ± 0.191 7 ) % respectively ( P 〈 0.05 ). The rate of apoptosis was (6. 932 0 ± 0. 638 0) % in MC group and(3. 021 0±0. 254 0)% in Sen group (P 〈0. 05). Bcl-2 expression was up-regulated while Bax expression was down-regulated in Sen group compared to those in MC group (P 〈 0.05). Conclusion Senqi Fuzheng injection can lower the AI and protect myocardial cells from MC by increasing Bcl-2 expression and decreasing Bax expression.
