摘要
目的观察颈交感神经阻滞(sympathetic block,SB)对烧伤小鼠早期炎症介质核因子-κB(NF-κB)及其辅活化子的影响,初步探讨SB在严重创伤中治疗作用的分子机制。方法将雄性昆明小鼠分为正常对照组、烧伤对照组、LSB组(烧伤后使用利多卡冈进行SB治疗)、RSB组(烧伤后使用罗哌卡因进行SB治疗),每组5只。烧伤为TBSA15%~20%m度。烧伤后6h采用Western blotting测定肝组织NF-κBP65、类同醇受体辅活化子-3(SRC-3)的蛋白表达及核转位。结果烧伤对照组肝组织NF-κBP65、SRC-3蛋白表达水平及核转位显著增加;与烧伤对照组相比,LSB组NF-κBP65蛋白表达无显著变化,SRC-3蛋白表达及NF-κBP65、SRC-3核转位显著下调;RSB组NF-κBP65、SRC-3蛋白表达及核转位均显著低于烧伤对照组,且比LSB组下调更明显。结论烧伤早期(6h)NF-κB及其辅活化子SRC-3蛋白表达水平及核转位显著上调,SB在严重创伤中的治疗作用可能是通过抑制NF-κB功能活性,降低炎症反应程度而实现的。
Objective To observe the effect of cervical sympathetic block (SB) on NF-κB and its coactivator during the early stage in burned mice. Methods Kunming male mice were randomly divided into normal control, burn control, LSB group (treated immediately by SB with Lidocaine after burn injury) and RSB group (treated immediately by SB with Ropivacaine after burn injury). Third degree burn injury involving 15% - 20% of TBSA was produced. At 6 h after burn, the expression and nuclear translocation of hepatic NF-κB P65 and steroid receptor coactivator-3 (SRC-3) were detected by Western blotting. Results After burn injury, the expression and nuclear translocation of hepatic NF-κB P65 and SRC-3 were markedly elevated as compared with normal control group. The level of SRC-3 and nuclear translocation of NF-κB P65 and SRC-3 in LSB group were significantly downregulated compared with that of burn control group, but the NF-κB P65 was no significant difference with that of burn control group. The expression and nuclear translocation of NF-κB P65 and SRC-3 in RSB group were obviously lower than burn control group and LSB group. Conclusion The expression and nuclear translocation of hepatic NF-κB P65 and SRC-3 are markedly elevated during the early stage of burn injury. SB can exert supression on the function of NF-κB and reduce the degree of systemic inflammatory response, which may be the mechanisms for therapeutic effect of SB on severe trauma.
