摘要
目的:利用脂质体转染肿瘤坏死因子受体2(TNFR2)于喉鳞癌裸鼠模型中,探讨转染的最佳剂量及时间。方法:构建喉鳞癌裸鼠模型,将不同剂量TNFR2于不同时间转染至模型中,利用流式细胞术及免疫组织化学法测定TNFR2的表达。结果:鳞状细胞癌荷瘤裸鼠的瘤组织基本不表达TNFR2。转染TNFR2后,48 h蛋白表达达高峰,主要为细胞膜表达。给瘤体内转染5μg TNFR2效果最佳,蛋白表达水平最高。结论:TNFR2的最佳转染剂量为5μg,48 h可达高峰。此结果为喉鳞癌的基因治疗提供了依据。
Objective:To establish the best dose and time of the transfection of tumor necrosis factor receptors 2 (TNFR2) in vivo. Methods: Flow eytometry and Immunohistochemistry were needed to measure the TNFR2 expression. Results: Results showed that 5 μg TNFR2 was the best dose and 48 h was the better transfection time. TNFR2 was not expressed in the empty vetor group. The membrane expression,which was positive,was at its peak 48 h after transfection Of TNFR2 and the transfection of 5μg TNFR2 showed positive results and the protein expression was at its highest peak. Conclusion:The best transfection dose of TNFR2 is 5 μg and can reach its peak in 48 h so it provides basic proof for the genetic treatment of the laryngeal squamous cacinoma.
