摘要
目的观察天麻钩藤饮对甲亢肝阳上亢证大鼠下丘脑蛋白质表达的影响,以期寻找差异表达的蛋白质。方法将30只大鼠随机分为3组,用灌服附子汤及L-T4的方法造成甲亢肝阳上亢证的模型,采用2D电泳分别分离各组下丘脑的总蛋白质,经考马斯亮兰染色,PDQuest 7.0软件分析发现差异蛋白,结合生物信息学对其进行质谱鉴定。结果模型组及治疗组与正常组相比,T3、T4明显增高(P<0.05)。3组蛋白斑点总体分布相似,主要分布在等电点pI3~8,分子量Mr14.4~75 kD,与正常组比较,模型组中蛋白质点表达上调的有18个,下调的点有24个;在模型组中上调的18个点中,与治疗组相比,其中有8个点表达下降,而模型组中下调的24个点中,与治疗组相比,其中有20个点表达上调。结合生物信息学进行质谱鉴定,发现其中6个蛋白质分别为硫氧还蛋白、血小板活化因子乙酰基水解酶IB-r亚单位、NSFL1辅助因子、延伸因子1亚型、微管蛋白β-5和NAD依赖脱乙酰基酶。结论天麻钩藤饮能调节下丘脑蛋白质的表达,提示天麻钩藤饮可能是通过对甲亢肝阳上亢证大鼠下丘脑蛋白质的影响而起到改善症状的作用。
Objective To observe the effect of Tianma Gouteng decoction on protein expression in hypothalamus in rats with hyperthyroidism in the type of Liver-yang hyperactivity and so as to search the differential protein. Methods The 30 rats were randomly divided into 3 groups to establish the models by drenching Aconiti Praeparatae Decoction and L-T4. The total proteins of hypothalamus were separated by two-dimensional gel electrophoresis(2-DE) and the differential protein was discovered after coomassie blue staining and PDQuest 7.0 software analysis and the mass-spectrometer was measured with biological information. Results The protein spots in 3 groups were mainly displayed at range of pi3-8, Mr(14.4-75)kD. Compared with normal control group, 18 spots of protein expression increased and 24 decreased in model group; compared with the treated group, 8 spots decreased within the increased 18 spots of model group, and 20 spots increased within the decreased 24 spots of model group. By massspectrometer, these proteins were Thioredoxin, Platelet-activating factor acetylhydrolase IB gamma subunit, NSFL1 cofactor p47, Elongation factl-beta, Tubulin beta 5 and NAD-dependent deacetylase sirtuin-2. Conclusions Tianma Gouteng decoction can accommodate the proteins of hypothalamus, which indicates that the decoction maybe act on the proteins to improve the symptoms of liver-yang syndrome in the hyperthyroidism.
