摘要
目的:研究高脂高糖饲养大鼠血浆游离脂肪酸和肝脏、肌肉甘油三酯沉积与胰岛素抵抗的关系.方法:健康雄性9周龄SD大鼠20只,随机分为实验组(n=10)高脂高糖饲料饲养和对照组(n=10)普通饲料饲养.实验6wk,比较2组大鼠血浆游离脂肪酸(FFA),血清胰岛素(FINS),空腹血糖(FPG),胰岛素抵抗指数(HOMA-IR)及肝脏甘油三酯(TG)异位沉积的情况.结果:在实验6wk与对照组比较,①FFA(mmoL/L,0.74±0.04vs0.62±0.05,P<0.01);②FINS(mU/L,31.96±2.58vs12.64±1.35,P<0.01);③FBG(mmoL/L,5.32±0.55vs5.12±0.47,P>0.01);④HOMA-IR(7.57±1.11vs2.87±0.35,P<0.01)均升高;⑤体内脂肪相对含量(g,8.81±1.27vs6.65±0.88,P<0.01);⑥油红染色病理切片计算机半定量分析,肝脏组织中TG含量高于正常对照组(99.5±4.7vs128.5±3.9,P<0.01),肌肉(127.6±3.1vs134.7±2.9,P<0.01)胰腺的TG沉积无明显差异(P>0.05).结论:高脂高糖饲养可以诱导大鼠胰岛素抵抗,其胰岛素抵抗的形成与FFA的增加及肝脏和肌肉组织TG异位沉积有关.
AIM: To explore the accumulation of triglyceride (TG) in liver, muscle and pancreas in high-fat-diet induced insulin resistance rat model and the relationship between the insulin resistance and the accumulation of TG in non-adipose tissues. METHODS: Twenty 9-week male SD rats were randomly divided into 2 groups and fed for 6 weeks with high-fat diets (high-fat group, n = 10) and standard diets ( control group, n = 10). Blood was collected every 7 d for detecting free fatty acids (FFA), fasting insulin (FINS) and fasting plasma glucose (FPG). Insulin resistance was determined with Homa-model analysis(HOMA-IR). The accumulation of TG in liver, muscle and pancreas was investigated by oil red O staining and computer semi-quantitative analysis on the 6th week. RESULTS: In highfat group as compared with control group, FFA (mmoL/L, 0.74 ±0.04vs0.62±0.05, P〈0.01) , FINS (mU/L, 31.96±2.58 vs 12.64 ±1.35, P〈0.01), FPG (mmoL/L, 5.32 ±0.55 vs 5.12 ± 0.47, P 〉 0.01 ), HOMA-IR (7.57 ± 1.11 vs 2.87 ± 0.35, P 〈0.01 ) were increased significantly . On the 6th week, the amount of TG accumulation in liver and muscle was significantly higher than that in pancreas. There was no significantly difference in the TG accumulation in pancreas between the highfat group and the control group ( P 〉 0.05 ) , but significantly in the liver(99.5 ±4.7 vs 128.5 ±3.9, P〈0.01) and muscle (127.6±3.1 vs 134. 7 ±2. 9, P 〈0.01). CONCLUSION: The high-fat-diet can induce insulin resistance in SD rats, which seems to be related to higher serum FFA and the accumulation of TG in liver and muscle.
