摘要
目的观察蛋白激酶Cδ抑制剂在6-OHDA诱导多巴胺能神经细胞系SH-SY5Y细胞凋亡过程中所起的作用,探讨帕金森病可能的分子发病机制。方法体外培养SH-SY5Y细胞,观察PKC抑制剂对6-OHDA毒性作用的影响,分别用台盼蓝染色、流式细胞仪和透射电镜法观察细胞凋亡。结果PKCδ抑制剂Rottlerin可减轻6-OHDA引起的细胞凋亡。而总PKC抑制剂Bisindolylmaleimide I(Bis)和钙依赖性PKC(α和β)抑制剂G 6976不能减轻6-OHDA引起的细胞凋亡。结论PKCδ抑制剂对6-OHDA诱导的SH-SY5Y细胞凋亡产生保护作用,PKCδ可能直接参与了帕金森病人的神经元凋亡过程。
Objective To observe the effect of phosphorylation protein kinase C delta(PKCδ) in the procedure of SH-SY5Y cell apoptosis induced by 6-OHDA and investigate the potential molecular pathogenesis of Parkinson's disease. Methods Hematoxylin-Eosine, trypanhlau staining and transmission electron microscope were applied to measure the contribution when dopaminergic SH-SY5Y cells exposed to the inhibitors of PKC before 6-OHDA for 24 hours. Results Rottlerin, the inhibitor of PKCδ, protected SH-SY5Y cells apparently. As contrast, Bisindolylmaleimide Ⅰ, the inhibitor of general PKC and Goe6976, the inhibitor of calcium-dependent PKC, had not protective role. Conclusion The inhibitor of PKCδ can reduce apoptosis of SH-SY5Y cells induced by 6-OHDA, PKCδ may directly participate in neurodegeneration process in Parkinsonian.
