摘要
目的探讨了硫酸铵对三种不同相对分子质量聚乙二醇双水相体系的分相能力和双水相体系对芦荟多糖的提取分离。方法以相对分子质量为6000的聚乙二醇/硫酸铵双水相体系从芦荟凝胶汁中提取分离得到芦荟多糖,以甘露糖为标准,采用浓硫酸-苯酚法对芦荟多糖的含量进行测定,并用红外(IR),紫外(UV)等手段对多糖的结构进行初步分析。结果确定以聚乙二醇6000的分相能力最好,聚乙二醇/硫酸铵双水相体系提取得到的芦荟多糖含量高于传统的醇沉法所得的芦荟多糖,醇沉法提取分离芦荟多糖的含量为68.39%,聚乙二醇/硫酸铵双水相体系为75.63%,而且聚乙二醇/硫酸铵双水相体系提取法大大的减少了有机溶剂的用量。结论PEG6000/硫酸铵双水相体系是一种很好的分离提取芦荟多糖的方法。
OBJECTIVE To research the phase separation ability of different PEG/( NH4 ) 2 SO4 system and extraction of aloe polysaccharides by aqueous two phase system. METHODS Aloe polysaccharides was extracted from aloe gel by the mean of PEG 6 000/ ( NH4 )2 SO4 aqueous two phase system. The contents of polysaccharides was determined by phenol-sulfuric acid method according to the standard of D-mannose, the structure of aloe polysaccharides was analyzed by IR, UV. RESUILTS PEG 6 000/( NH4 ) 2 SO4 aqueous two phase system was chosen for its best phase separation ability, more aloe polysaccharides was extracted by using PEG 6 000/ (NH4)2SO4 aqueous two phase system (75.63%), compared to ethanol-precipitated(68.39% ), which more organic solvent was used. CONCLUSION PEG 6 000/( NH4 ) 2 SO4 aqueous two phase system is a good method for extracting aloe polysaccharides.
