摘要
本文旨在考察铁对小鼠肝脏铁水平和TfR(transferrrin receptor)mRNA的影响,研究二者的关系。试验1选取40只昆明白雌鼠,随机分为5组,每组8只。对照组(处理1)饲喂不添加任何铁源的基础饲料,试验组(处理2、3、4、5)的饲料中分别添加硫酸亚铁(Fe60mg/kg)、硫酸亚铁(Fe120mg/kg)、甘氨酸铁(Fe120mg/kg)、甘氨酸铁+硫酸亚铁(Fe120mg/kg)。40d后处死取肝脏;试验2选取对照组子一代,不同铁剂处理,3d后处死取肝脏。测定肝脏铁含量;用相对定量RT-PCR方法,以β-actin为内标,测肝脏TfR mRNA丰度。结果表明:(1)试验1中,饲粮中添加不同来源和不同水平的铁能显著影响肝脏铁含量和TfR mRNA基因表达水平,处理组5较对照组相比差异极显著(P<0.01);(2)试验2中,子一代添加甘氨酸铁组和硫酸亚铁组间TfR mRNA基因表达水平差异不显著(P>0.05);(3)鼠肝脏铁含量与TfR mRNA的表达量成强负相关。铁可显著影响肝脏铁含量和TfR mR-NA表达量,二者成负相关。
To investigate the effect of iron on iron content and transferrin receptor (TfR) mRNA and the relation between them of mice livers. Single tube relative-quantitative RT-PCR method was used and β-actin as a internal standard. Forty Kunming White female mice were selected and divided randomly into 5 groups,raised according to normal nutrition standard of experimental animal with different source and dose of iron for 40 d. Group 1 (control)mice were fed purified diets (without supplement iron) and group2、3、4、5 were fed diets with supplement iron of different source and dose:FeSO4 (60 mg/kg Fe) ; FeSO4 (120 mg/kg Fe) ;Fe-Gly(120 mg/kg Fe) ;1/2 FeSO4 + 1/2 Fe-Gly(120 mg/kg Fe) ,respectively. In addition,feeded the offspring of control for 3 d with FeSO4 (120 mg/kg Fe) and Fe-Gly (120 mg/kg Fe),respectively. Slaughter all the mice and removed livers for total RNA extraction and iron quantification. The result showed 1) after trial 1 ,different iron source and dose can increased the iron level very significantly(P〈0.01);at the same time iron can decreased TfR mRNA level significantly(P〈0.05) ,especially in treatment 5,2) in trial 2,the result was not significant(P〉0.05). Fe-Gly can increased iron level significantly and decreased the expression level of TfR mRNA. The relation between TfR mRNA and iron level was inverse,
