摘要
目的建立大鼠鼠胚海马神经元原代培养的方法。方法取孕龄17-18天的Wistar大鼠腹中胎鼠,血清培养法进行海马神经元的原代培养,然后用倒置显微镜进行形态学观察和神经元特异性烯醇化酶免疫组织化学方法进行神经元的鉴定。结果培养8天后的海马神经元经形态学和组织化学方法观察证实神经细胞发育成熟。结论大鼠鼠胚海马神经元原代培养是一种新型的培养方法,具有明显的优势。
Objective : To establish primary culture method for hippoeampal neurons of embryo of rat. Methods : Embryo of rat which was pregnant for 17 - 18 days was used in experiment. Hippocampal neurons were cultured with Serum combination. Cultured neurons were observed by inverted microscope and identifieated by the method of NSE. Results: Hippocampal neutons were founded to be maturated by the methods of morphology and try. Conclusions : Primary culture method for hippocampal neurons of embryo of rat is a new method of culture , which is proved to outweigh other methods.
