外源性IFN-γ对植入前期妊娠大鼠下丘脑内IL-4阳性细胞的影响

The Effect of Exogenous IFN-γ on the Expression of IL-4 Immunoreactive Cells in Hypothalamus of Pregnant Rat in Preimplantation

摘要 为了解外源性IFN-γ对植入前期大鼠下丘脑内IL-4表达的影响,通过阴道口肌肉注射不同剂量IFN-γ后观察植入前期大鼠下丘脑内IL-4表达的变化,并且用免疫组织化学SP法对其检测。结果表明:在下丘脑的视前大细胞核(POMA)、视前内侧核(POM)、交叉上核(SO)、视上核(SC)、室旁核内侧部(PVM)和室周核(HPV)等处均有IL-4阳性细胞的表达分布;试验1组(注射IFN-γ25 000 IU)和试验2组(注射IFN-γ75 000 IU)各个核团中阳性细胞数与对照组相比较少而且差异显著(P<0.05),2个试验组相比,在部分核团中阳性细胞数试验1组高于试验2组,差异显著(P<0.05)。结果说明下丘脑内IL-4阳性细胞随着外源性IFN-γ剂量的增加而减少,从而说明外源性IFN-γ有可能会影响下丘脑中IL-4蛋白的表达分布。 The model of pregnant sprague-dawley rat in preimplantation was established to study the expression and distribution IL-4 immunoreactive cells in the hypothalamus after injected exogenous IFN—γ in muscle of vagina by using ultrasensitive immunohistochemical SP method. The results were as follows. IL-4 immunoreactive cells were found in nucleus pre-opticus magno cellularis, pre-opticus medialis, nucleus suprachiasmaticus, nucleus supra-opticus, nucleus paraventricularis medialis, nucleus periventricularis hypothalami and so on. There are more IL-4 immunoreactive cells in control group (normal sodium), compared with the first experiment group (injected with IFN—γ 25 000 IU) and the second experiment group (injected with IFN—γ 75 000 IU). IL-4 immunoreactive cells in the first experiment group were more than the second experiment group. The data presented indicated that IL-4 immunoreactive cells in hypothalamus were decreased as the dose of exogenous IFN—γ increased, and it also showed us that exogenous IFN—γ could effect the expression of IL-4 cytokine in hypothalamus.
