摘要
利用BlastX将球毛壳菌(Chaetomium globosum)ESTs序列与GenBank的Nr数据库进行相似性搜索,获得含有细胞色素c氧化酶亚基5基因(cox5)全长cDNA序列的EST(BP099084),以此设计引物。以球毛壳菌总RNA反转录产物为模板进行PCR扩增,获得球毛壳菌cox5的cDNA序列。cDNA全长685bp,编码169个氨基酸的多肽,蛋白质分子量18.3KD。蛋白质家族预测其属于cox4家族。球毛壳菌cox5是具有导肽的跨线粒体内膜的蛋白质。BlastP相似性分析表明cox5氨基酸序列保守性不高,与其相似性最高的是柄孢霉的cox5(Podospora anserina,CAB76570),相似性为75%。cox5基因的cDNA序列及推测的氨基酸序列在GenBank上登录(登录号分别为DQ082750、AAY85812)。
The EST representing the full-length cDNA sequence of cytochrome c oxidase subunit V gene ( cox5 ) was obtained by blastx searching EST from Chaetomium globosum against Nr database of GenBank. According to the sequence of EST representing cox5, primers for PCR amplifying cox5 gene were designed. PCR was conducted using the template reversely transcribed from total C. globosum RNAs, and a length of 685 bp full-length cDNA of cox5 was gained. The cox5 gene encodes the protein of 169 amino acids with molecular mass of 18.3 KD. Protein family prediction indicates that this protein belongs to cox4 family, cox5 is a transmembrane protein located in the mitochondrion's inner membrane with leader sequence. BlastP analysis reveals that amino acid sequences of cox5 genes from some fungal species share low similarity. The cox5 from C. globosum shares the highest similarity with that from Podospora anserine, and the similarity is 75%. The cDNA and deduced amino acid sequence of cox5 gene were accepted by GeneBank (accession numbers: DQ082750, AAY85812).
