摘要
目的通过霉酚酸酯(MMF)干预UUO大鼠模型,动态观察单核细胞趋化因子(MCP-1)、TGF-β1在梗阻侧肾组织中的表达。方法54只雄性SD大鼠随机分为假手术组、UUO模型组和MMF治疗组。于术后第7天、14天、21天每组分别处死6只大鼠。免疫组织化学染色观察MCP-1和TGF-β1在肾间质中的表达。结果在正常肾间质中,MCP-1和TGF-β1均只有少量表达,两者在UUO模型组中的表达均显著增高,MMF治疗组各时间点与相应UUO模型组比均有不同程度降低。结论MMF能部分延缓UUO模型肾间质纤维化的进展,可能与其早期下调MCP-1在肾间质中的表达抑制炎症反应,进而抑制致纤维化的关键因子TGF-β1表达有关。
Objective To observe the expression of MCP-1 and TGF-β1 in the obstructive renal tissue of UUO rats intervened by MMF. Methods Fifty-four male Sprague-Dawley rats were randomly divided into three groups: sham-operated rats group, UUO model group and MMF-treated group. Six rats of each group were sacrificed at the 7th, 14th, 21st day after operation. The expression of MCP-1 and TGF-β1 in renal interstitium were detected with immunohistochemistry. Result The expression of MCP-1 and TGF-β1 were at low level in normal renal interstitium. In group UUO, MCP-1 and TGF-β1 were obviously higher than those in group SOR. MCP-1 and TGF-β1 were decreased after treating by MMF at each time point. Conclusion In UUO rats, the possible mechanism with which MMF prevents renal interstitial fibrosis is that MMF may inhibit inflammation reaction by down regulating MCP-1, and then inhibit TGF-β1.
