摘要
目的研究抗凋亡基因Bag-1和Bcl-2在结肠癌组织中的表达及其意义,并探讨两者间的关系。方法应用免疫组织化学SABC法(链霉亲合素-生物素-过氧化物酶复合物法)对64例结肠癌组织及10例正常结肠组织中Bag-1和Bcl-2基因的表达进行检测。结果Bag-1和Bcl-2在结肠癌中的表达与结肠癌病理类型、肿瘤直径、浸润深度及有无淋巴结转移无关,但Bag-1与病理分级、远处转移、Dukes分期及预后密切相关(P<0.05),而Bcl-2与这几项临床病理因素无明显相关性(P>0.05);Bag-1和Bcl-2在结肠癌的发生发展过程中呈正相关(r=0.475)。结论结肠癌组织中有不同水平的Bag-1和Bcl-2蛋白的高表达,它们可以作为结肠癌早期筛选的一项指标,并对疾病的预后有着重要意义。
[Objective ] To investigate the expression and significance of anti-apoptotic gene Bag-1 and Bcl-2 in colon cancer, and evaluate their relation. [Methods] The expression of the Bag-1 and Bcl-2 in 64 colon cancer tissue and 10 normal colon tissue samples was examined by immunohistochemical technique (sptreptavidin-biotin- peroxidase complex method). [Result] The expression of Bag-1 in colon cancer was closely correlated with pathologic grade, distante metastasis, Dukes stage and prognosis had no effect on the pathologic type, tumor diameter. depth of invasion and lymphoid node metastasis of the cancer. However, Bcl-2 had no significant correlation with all these clinical and pathologic factors. We also found that there was a positive correlation between Bag-t and Bcl-2 in the developing progress of the colon cancer. [Conclusion] There is high expression of Bag-1 and Bcl-2 protein in colon cancer. They might be regarded as a biomarker for the diagnosis of early period colon cancer, and have significant meaning for the prognosis of colon cancer.
