摘要
基于PCR技术的SSR标记已经广泛用于基因鉴定、遗传图谱和分子标记辅助育种。为了建立一种适于SSR-PCR的DNA快速提取方法,利用一种新的磨样技巧和减少DNA提取步骤的方法,以大豆叶片为样品对传统CTAB法进行改进。结果表明:新的提取方法与传统CTAB法相比,具有提取速度快、试剂费用低和不需在液氮中磨样等优势。提取的DNA可用于大豆SSR-PCR分析。
SSR marker, a PCR- marker assisted selection. To CTAB method was improved based marker, has been widely used to identify and tag gene as well as in develop a faster DNA extraction method for SSR-PCR, the conventional by a novel pulverizing technique to simplify extractive process in soybean leaves. The results showed that new extraction had advantages of being faster, less expensive and no re quiring liquid nitrogen to pulverize the samples than conventional CTAB method, and the DNA derived from new method could be used to SSR-PCR analysis in soybean.
