硒对Ox-LDL诱导牛内皮细胞Caspase-3酶的作用

摘要 目的探讨硒对氧化型低密度脂蛋白(Ox-LDL)诱导体外培养牛主动脉内皮细胞Caspase-3酶的影响。方法Ox-LDL与硒处理牛主动脉内皮细胞后,通过流式细胞仪、DNA电泳检测凋亡细胞,测定Caspase-3酶活性。结果Ox-LDL作用内皮细胞后,细胞凋亡DNA电泳呈梯度条带;加入硒干预后,内皮细胞平均凋亡率及Caspase-3酶活性吸光度比值较Ox-LDL组明显降低(P<0.05)。结论硒能减少Ox-LDL诱导体外培养的牛主动脉内皮细胞凋亡,部分通过抑制Caspase-3酶活性而发挥抗凋亡的细胞保护效应。 Objective To investigate the effects of antioxidant selenium on the oxLDL - induced apoptosis of bovine aortic endothelial cells(BAECs) and relevant mechanism. Methods BAECs were isolated and cultured, The endothelial cells were cultured with selenium for 12 hours and then were treated with oxLDL for 24 hours. The apoptosis of the endothelial cells was checked by the flow cytometry and DNA electrophoresis, while caspase - 3 activity was measured simultaneously. Results The DNA electrophoresis showed typical ladders. While the endothelial cells cultured with selenium, the mean of apoptotic rates and caspase - 3 enzyme activity significantly decreased (P 〈 0. 05). Conclusion Selenium can reduced the apoptosis of cultured endothelial cells induced by oxLDL. This effect may be mediated by significantly suppressing caspase - 3 activity.
