摘要
目的观察p38丝裂原活化蛋白激酶(p38 MAPK)抑制剂SB203580对大鼠膝骨关节炎(OA)的软骨保护作用。方法将40只大鼠随机均分为A、B、C、D组,均行单侧膝关节前交叉韧带切断。A、B组术后分别于关节腔注射0.1ml浓度为100、10μm/L的SB203580,每周1次,连续6周;C组注射等量生理盐水;D组不做任何处理。术后6周处死动物,比较各组软骨的大体变化、OA评分、软骨表面分级及Mankin炎性分级。结果A、B组软骨大体评分、软骨表面分级及Mankin炎性分级均轻于C、D组。结论关节腔注射p38 MAPK抑制剂能延缓软骨退变程度,具有软骨保护作用。
[Objective] TO observe the protective effect of p38 mitogen activated protein kinase inhibitor SB203580 on cartilage of experimental rats osteoarthritis (OA). [Methods] 40 SD rats underwent unilateral anterior cruciate ligament transection (ACLT).and were randomly divided into A,B,C,D groups. Rats of group A and B received 0. 1 ml intra-articular injection of SB203580 at 100, 10 μm/L after surgery immediately for 6 weeks, once a week. Intra-articular of those in group C received 0.1 ml 0.9% sodium chloride ,group D were not injected as control. On the 6th week after surgery,the rats were sacrificed. The cartilage changes,OA grade,cartilage surface class and Mankin inflammation class were compared. [Results] The cartilage changes,cartilage surface class and Mankin inflammation class in group C and D were more severe than those in experimental group A,B. [Conclusion] It is suggested that p38MAPK inhibitor SB203580 might reduce the severity of cartilage degeneration and have the chondroprotective effect.
