摘要
目的检测同种异体组织工程骨植入小香猪皮下后白细胞介素2(IL-2)及肿瘤坏死因子α(TNF-α)的表达,探讨同种异体骨髓间充质干细胞(MSCs)作为种子细胞构建组织工程骨的可行性。方法猪骨髓间充质干细胞体外诱导分化为成骨细胞,与猪源生物衍生骨材料复合培养,构建组织工程骨,植入15头免疫功能健全的异体小香猪背侧脊柱旁左侧皮下作为实验组,对侧植入单纯的脱钙骨基质(DBM)作为对照,于术后1、2、4、8和12周抽取静脉血,并行双侧局部组织取材,组织块作低温匀浆,ELISA法检测IL-2及TNF-α的水平变化。结果实验组和对照组术后各时间点IL-2及TNF-α水平无统计学差异,IL-2及TNF-α水平在术后第2周开始增高,第2~4周维持高水平,8周后随时间的延长而下降。外周血无明显变化。结论小香猪MSCs和DBM复合构建组织工程骨,植入异体小香猪体内引发微弱的免疫反应,同种异体MSCs可选择作为构建骨组织工程的种子细胞。
Objective To explore the feasibility of allogeneic mesenchymal stem cells (MSCs) as seed cells to construct tissue engineered bone by detecting the expressions of IL-2 and TNF-α to Chinese minipig strain of Xiangzhu after implanting these tissue engineered bones. Methods Engineered bones were constructed with osteoblasts which derived from allogeneic MSCs and DBM in vitro,and then were implanted to subcutanenous tissue in left back as experimental group,DBM only were implanted to right as control group. Every 3 minipig strain of Xiangzhu were sacrificed in 1,2,4,8,12 week postoperatively and the expressions of IL-2 and TNF-α in blood and graft samples were detected quantitatively by enzyme-linked immuneosorbent assay (ELISA). Results There was no significant difference in the contents of IL-2 and TNF-α between two groups. The contents of IL-2 and TNF-α increased from 2 week and maintained high level from 2--4 weeks,but decreased after 8 weeks. Conclusion Tissue engineered bones constructed with allogeneic MSCs and DBM show low immunogenicity. Allogeneic MSCs may be used as seed cells to construct tissue engineered bone.
