摘要
目的观察目前临床常用的治疗骨关节炎(OA)药物对骨关节炎动物模型关节液中细胞因子IL-1β、TNFα水平的影响。方法按照ACLT法建立兔膝关节骨关节炎模型,30只实验动物随机等分成6组:(1)透明质酸钠组,(2)硫酸氨基葡萄糖组,(3)塞来昔布组,(4)独活寄生汤组,(5)盐水组,(6)空白组。各治疗组分别给予药物治疗4周,然后抽取关节液,ELISA法检测关节液中IL-1β、TNFα的含量。结果透明质酸钠、硫酸氨基葡萄糖和塞来昔布组关节液中IL-1β、TNFα水平均显著低于对照组(P<0.01);独活寄生汤组关节液中IL-1β、TNFα水平均与对照组无显著性差异(P>0.05);透明质酸钠组模型关节液中IL-1β、TNFα水平均显著低于硫酸氨基葡萄糖和塞来昔布组(P<0.05)。结论四种药物中透明质酸钠、硫酸氨基葡萄糖和塞来昔布均能够抑制OA模型关节滑液中IL-1β、TNFα的水平,其中透明质酸钠更加显著。
Objective To determine the effect on four current drugs on influencing synovial fluid IL-1β and TNFα levels of OA model, Methods Experimental OA models of rabbit knee joint were setup according to ACLT method. 30 model were randomly divided into 6 groups and every group had 5 animals, Those groups were sodium hyaluronate group, glucosamine sulfate group, celecoxib group, duhuojisheng group, saline group and control group, All treatment groups were received medications for 4 weeks, The IL-1β and TNFα levels in synovial fluid were measured by ELISA method. Results The synovial fluid IL-1β and TNFα levels of sodium hyaluronate group, glucosamine sulfate group and celecoxib group are significantly lower than the control group (P 〈 0.01). The levels of IL-1β and TNFα of duhuojisheng group have not significantly difference with control group (P 〉 0.05), The levels of IL-1β and TNFα of sodium hyaluronate group are significantly lower than that of glucosamine sulfate group and celecoxib group (P 〈 0.05), Conclusions Sodium hyaluronate, glucosamine sulfate and celecoxib could significantly decrease the levels of IL-1β and TNFα in synovial fluid of OA model.
