摘要
目的:构建脱细胞猪主动脉瓣膜支架,为体外构建组织工程心脏瓣膜提供适宜的支架材料。方法:新鲜猪主动脉瓣叶,采用“低渗液-0.5%SDS-核酸酶”处理,进行大体、光镜、电镜观察和DNA含量测定。结果:“低渗液-0.5%SDS-核酸酶”可以完全脱去瓣膜细胞,同时较好地保持了胶原纤维和弹力纤维的原有排列和形态。结论:本方法可以成功构建脱细胞猪主动脉瓣膜支架,可进一步用于体外构建组织工程心脏瓣膜的研究。
Objective: To construct an acellular valve scaffold from porcine aortic valves for tissue engineering of heart valve. Methods: A fresh porcine aortic valve leaflet was decellularized by "low osmotic pressure of Tris buffer-0.5%SDS-RNase and DNase", and assessed by histology, scanning electron microscopy, and the levels of DNA content to confirm the removal of cells. Results: The cells of porcine aortic valve leaflet were removed effectively by "low osmotic pressure of rids buffer-0.5%SDS- RNase and Dnase" methods. The collagen and elastin were kept intact. Conclusion: An acellular valve scaffold can be obtained successfully through this procedure.
