糖尿病大鼠肾小管整合素连接激酶的表达及氯沙坦的干预作用 被引量:7

Expression of Integrin-Linked Kinase in Renal Tubular Epithelial Cells in Diabetic Rats and the Effect of Losartan on it

摘要 目的:观察糖尿病大鼠肾小管上皮细胞整合素连接激酶(ILK)的表达以及氯沙坦对其的影响。方法:雄性Wistar大鼠腹腔内注射链脲佐菌素(STZ)诱导糖尿病模型,随机分为模型组和氯沙坦组;另设正常组腹腔注射缓冲液。第8周、第16周测24h尿蛋白定量和血肌酐,处死大鼠,取肾组织行HE、Masson染色,观察肾脏病理改变;免疫组化法检测大鼠肾小管上皮细胞ILK、TGF-β1的表达。结果:模型组大鼠肾小管上皮细胞胞浆ILK表达增高,与小管间质病变程度一致;同时TGF-β1表达亦增高,并与ILK表达呈正相关性。氯沙坦组大鼠尿蛋白较模型组明显减少,肾小管间质病变减轻,肾小管上皮细胞ILK、TGF-β1表达明显减弱。结论:糖尿病大鼠肾小管上皮细胞存在有ILK的高表达,ILK的表达强度与肾小管间质损伤程度相一致,并与TGF-β1的表达呈正相关;氯沙坦能下调肾小管上皮细胞ILK的表达。 Objective:To investigate the expression of ILK in renal tubular epithelial cells in diabetic rats;To observe the effect of Losartan on ILK expression in diabetic rats. Methods:Rat models of diabetic mellitus were established by intraperitoneal injection of streptozotocin, and those models were randomly divided into two groups after these diabetic mellitus models were successful established: diabetes without therapy group and diabetes therapy group with Losartan. The control group were only injected buffer solution. Eight weeks and sixteen weeks after therapy with Losartan, urine was collected for 24 hours to measure 24 - hours proteinuria and the serum creatinine concentration. Kidneys were examined at 8wk and 16 wk by light microscopy, and the expression of ILK and TGF- β1 was detected by immunohistochemistry analyses. Results: ( 1)The renal tubular injury and interstitial lesion of rats in diabetes group presented as degeneration, necrosis , inflammatory cell infiltration , and interstitial collagen deposition. (2)The expression of. ILK and TGF - β1 increased during the process of injury of tubular epithelial cells in diabetes group. The level of expression of ILK correlated positively with that of TGF-β1; In contrast, normal tubular epithelium in control failed to express ILK and TGF- β1. (3)Losartan down- regulated the expression of ILK and TGF- β1 in diabetes therapy group. Conclusion: (1) The overexpression of ILK occured in proximal tubular epithelial cells in the process of renal tubulointerstitial fibrosis in diabetes Rats. (2)The role of ILK in tubulointerstitial injury and fibrosis in diabetic nephropathy maybe correlate with TGF- β1 (3)Losartan restraining the expression of ILK is a possible mechanism of its protective effect on diabetic nephropathy.
