自身免疫性心肌炎小鼠心肌Annexin Ⅵ的表达及意义

摘要 目的:研究膜联蛋白Ⅵ(annexin Ⅵ)在自身免疫性心肌炎小鼠心肌中的表达变化,探讨其在心肌炎各阶段心功能改变中的意义。方法:人工合成多肽诱导BALB/c小鼠自身免疫性心肌炎;分别于免疫后第14,21,30,60,90,120天测心身比值,HE染色观察心肌炎症情况,免疫组化与Western blot法观察annexin Ⅵ在心肌组织中的分布与表达情况。结果:在心肌炎的炎症慢性期心肌细胞的胞浆与间质中均可表达annexin Ⅵ;与正常对照组相比,急性期annexin Ⅵ表达减少(P<0.05),随炎症迁延进展慢性期表达逐步增加(P<0.01)。结论:人工合成的多肽可以成功诱导出自身免疫性心肌炎;炎症早期annexin Ⅵ表达下降而晚期表达增加,可能参与了心功能代偿与失代偿期的变化。 Objective: To research the alteration of annexin Ⅵ in myocardium of mice with autoimmune myocarditis, and to study the effect of annexin Ⅵ on myocardial contractility. Methods: BALB/c mice were immunized by the emulsion of artificially synthesized polypeptides which derived from cardiac myosin heavy chain and CFA (complete Freund' s adjuvant). Body weight and heart weight were recorded for the calculation of the heart/body ratio HE-staining was used to identify the degree of inflammation, immunohistochemistry and Western blot were used to observe the distribution and content of annexin Ⅵ at 14, 21, 30, 60, 90 and 120 d separately after immunization. Results: Annexin Ⅵ was expressed both in cytoplasm and interstitium in the chronic phase; compared with control group, the content of annexin Ⅵ decreased in acute phase, and increased in chronic phase. Conclusion: The polypeptides could induce myocarditis. The decreasing of annexin Ⅵ in the earlier period and the increasing of annexin Ⅵ in the advanced stage of inflammation probably take part in the changes of cardiac function.
