摘要
目的 观察小鼠烧伤后早期类固醇受体辅活化子(SRC)蛋白表达的时相变化,初步探讨烧伤早期糖皮质激素抵抗的分子机制。方法 将30只健康C57/129雄性小鼠随机分为正常对照组与烧伤组,烧伤组给予背部TBSA15%~20%Ⅲ°烧伤。分别于伤后1、6、12、24小时采用Westernblot测定肝、脾组织SRC家族(SRC-1、GRIP-1、NC0A-3)蛋白表达水平的变化。结果 烧伤组肝组织SRC-1蛋白表达水平在烧伤后l小时即显著降低,6小时降至最低,以后逐渐恢复,至24小时与正常对照组相比无显著差异;正常脾组织SRC-1蛋白表达水平较低,测定结果无统计学意义。烧伤组肝组织GRIP-1、NC0A-3蛋白表达水平在烧伤后显著升高,12小时达到高峰,24小时仍显著高于正常对照组;烧伤组脾组织GRIP-1、NC0A-3在烧伤后1小时显著升高,GRIP-1在烧伤后6小时达到高峰,24小时与正常对照相比差异不显著;而NC0A-3在烧伤后12小时达到高峰,24小时显著高于正常对照组。结论 小鼠烧伤后早期SRC-1显著降低,GRIP-1、NC0A-3显著升高,烧伤后糖皮质激素抵抗的发生发展可能与SRC家族成员表达变化有关。
Objective To observe the changes of expression of steroid receptor coactivators (SRC) during the early stage in the burned mice,and the mechanism of glucocorticoid resistance(GCR). Methods Thirty C57/129 male mice were divided randomly into the control and the bum groups. After clipping of the hair, 15% - 20% thickness Ⅲ°bum injury was produced. At 1,6,12,24h after bum,the levels of hepatic and splenic steroid receptor coactivators( SRC-1 ,GRIP-1 and NCoA-3 ) were detected by western blot. Results The level of hepatic SRC-1 was markedly down-regulated lh after injury,and reached the lowest point 6h after injury,and gradually returned to the normal level 24h after bum injury. The expression of splenic SRC-1 of the control group was a little lower,which was not statistically significant. The hepatic GRIP-1, NCoA-3 in the burned mice was significantly elevated compared with that of the control group,and peaked 12h after injury. The splenic GRIP-1 ,NCoA-3 was markedly elevated lh after injury,and GRIP-1 was peaked 6h after injury,and gradually returned to the normal level,but NCoA- 3 was peaked 12h after bum,and still obviously higher than that of the control group 24h after bum. Conclusion The SRC-1 was markedly down-regulated,and the GRIP-1 ,NCoA-3 was markedly elevated during the early stage of bum injury,which indicates that the glucocorticoid resistance after bum injury may be associated with the changes of expression of steroid receptor coactivators.
