摘要
目的观察银杏叶提取物(GBE)对门静脉高压症大鼠肺血管核转录因子-κB(NF-κB)和α-平滑肌肌动蛋白(α-SMA)表达的影响。方法应用免疫组织化学染色方法检测普通雄性Wistar大鼠100只(将实验动物随机分为4组:(1)PHT组(门静脉高压症组);(2)GBE+PHT组(腹腔注射GBE);(3)对照组即生理盐水NS+PHT组(腹腔注射生理盐水);(4)假手术组。)中NF-κBp65、α-SMA的表达。结果假手术组中两者几乎无阳性表达,分别为0.129±0.001(0.229±0.101)。其余3组以PHT组和NS+PHT组表达最强,分别为0.329±0.004(0.349±0.089)、0.324±0.004(0.334±0.084),组间比较差异无统计学意义(P>0.05)。GBE+PHT组阳性表达较弱为0.198±0.003(0.288±0.103),与PHT组间比较差异有统计学意义(P<0.05)。结论门静脉高压可以引致全身多器官血管病变,NF-κB、α-SMA可能在其发病机制中起到重要的作用。
Objective To investigate the effect of gingkobiloba extract (GBE) on the expression of NF-κBp65 and α-SMA in pulmonary vessels of rats with portal hypertension (PHT). Methods Male Wistar rats (n = 100) were randomly allocated to 4 groups:( 1 ) portal hypertensive animals (PHT) ; (2) PHT in the presence of GBE ( PHT + GBE) ; (3) PHT in the presence of NS ( PHT + NS) ; (4) sham- operated animals. The expression of NF-κBp65 and α-SMA was detected by immunohistechemistry. Results The expression levels of NF-κBp65 and α-SMA in pulmonary vessels of sham operated animals were (0. 129 ±0.001) and (0.229 ±0. 101) respectively. In PHT group and iNS +PHT group,the expression levels of NF-κBp65 and α-SMA were 0.329 ±0. 004and 0.349 ±0. 089 ,and 0. 324±0.004 and 0.334± 0.084 respectively with no significant difference between the two groups ( P 〉 0.05). The expression levels of NF-κBp65 and α-SMA were lower in GBE + PHT group (0. 198 ±0.003 and 0. 288 ±0. 103 respectively) than in PHT group ( P 〈 0.05 ). Conclusion Portal hypertension may cause multiple organ vasculopathy. NF-κB and α-SMA may play an important role in portal hypertensive vasculopathy.
