姜黄素对人肝细胞HepG2中低密度脂蛋白受体基因表达影响的研究 被引量:9

摘要 目的研究不同浓度姜黄素对人肝细胞HepG2中内源低密度脂蛋白受体(LDLR)表达的影响,在受体水平上探讨姜黄素的调脂作用机制。方法以荧光试剂标记配体法,利用流式细胞仪技术和荧光显微镜技术研究姜黄素对HepG2细胞LDLR表达活性的影响。利用细胞免疫技术、结合荧光显微镜技术流式细胞术研究姜黄素对HepG2细胞LDLR表达数量的影响。结果姜黄素可显著增加人肝细胞HepG2内源LDLR的表达(P<0.05)。结论姜黄素能够通过增加人肝细胞HepG2LDLR的表达起到调节血脂和抗动脉粥样硬化的作用。 OBJECTIVE To investigate the influence of curcumin on the endogenous low density lipoprotein receptor (LDLR) expression in the human hepatoma cell line HepG2 and seek for the lipids-regulating mechanism of cureumin. METHODS The ligands of LDLR and LDL were labeled by a fluorescent reagent and DiI. The effect of curcumin on the expression of functional LDLR in HepG2 cell line was measured by Fluo-Microscopy(FM) and Fluorescence Flow Cytometric Methods(FFCM). The effect of curcumin on the numbers of LDLR expression in the HepG2 was measured by FM, ELISA and FFCM. RESULTS Cureumin obviously up-regulated the expression of LDLR in HepG2 (P 〈 0. 05). CONCLUSION Curcumin is a potent activator of LDLR expression. It may be one of its mechanisms to modulate the plasma LDL-C by increasing the expression of LDLR in Hepatocells.
