降脂益肝冲剂治疗非酒精性脂肪性肝炎的实验研究 被引量:2

摘要 目的探讨降脂益肝冲剂对高脂饮食诱导的大鼠非酒精性脂肪性肝炎模型的影响。方法24只雄性Wistar大鼠随机分为3组(每组8只):正常组给予普通饲料喂养,模型组和治疗组给予高脂饮食喂养。治疗组在高脂饮食12w后给予降脂益肝冲剂,同时模型组和正常组分别给予等量的生活饮用水灌胃,18w末处死各组大鼠。测定空腹血糖(FBG)、空腹胰岛素(FINS),计算胰岛素敏感指数(ISI);测定血清丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、总胆固醇(TC)、甘油三酯(TG),肝组织TC、TG、丙二醛(MDA)、超氧化物歧化酶(SOD)活性;HE染色观察肝组织病理变化。结果模型组大鼠血清转氨酶、血脂、肝组织TC、TG、MDA升高,SODI、SI降低,伴典型的脂肪性肝炎。模型组血清和肝组织TG、TC分别为(0.88±0.063)mmol/L和(1.20±0.04)mmol/L、TC分别为(3.81±0.57)mmol/L和(3.32±0.55)mmol/L;治疗组血清和肝组织TG、TC分别为(0.78±0.062)mmol/L和(0.52±0.05)mmol/L(、2.95±0.31)mmol/L和(1.63±0.20)mmol/L。模型组和治疗组ALT分别为(110.5±25.7)U.L-1和(77.5±16.1)U.L-1、AST分别为(273.5±27.5)U.L-1和(226.2±17.8)U.L-1,与模型组相比,治疗组的各项指标都有所改善,差异有统计学意义。结论降脂益肝冲剂能通过调节脂质代谢,改善胰岛素抵抗,对抗氧化应激和脂质过氧化损伤,降低肝脏的脂质沉积有效地治疗大鼠非酒精性脂肪性肝炎。 Objective To explore the therapeutic effect of Jiangzhi Yigan Chongji on nonalcoholic steatohepatitic rats chronically fed with high-fat diet. Methods Twenty four male Wistar Rats were randomly divided into 3 groups (8 rats each group):normal group fed with standard diet, model group and treated group fed with high-fat diet,the treated group was given Jiangzhi Yigan Chongji (1ml/100g,its concentration is 0.8g/ml,twice every day)by intragastric administration after 12 weeks, meanwhile,the normal and model group were separately given life drinking water by the same means after 18 weeks, they were put to death. Results The model rats developed hyperlipidemia,increase of aminotransferase in surum, total cholesterol(TC),triglyeride (TG),malondialdehyde (MDA)in liver and decrease of the vitality of superoxide dismutase(SOD) in liver with steatohepatitis shown in hepatic histopathology.Compared with the model group, the treated group shown improvement of every data,and with statistical difference. Conclusion Jiangzhi Yigan Chongji has satisfactory therapeu- tic effects on seatohepatitic induced with high-fat diet. Regulating lipid metabolism ,improving insulin resistance,resisting oxidative stress and lipid peroxidation damage,decreasing lipidoses in liver may be some of its therapeutic mechanisms.
