摘要
目的探讨极低密度脂蛋白(VLDL)、氧化型极低密度脂蛋白(OX-VLDL)及肿瘤坏死因子(TNF-α)对正常人肝细胞增殖及三者对肝细胞凋亡的影响。方法将不同浓度VLDL,OX-VLDL及TNF-α分别与正常人肝细胞(HL-7702细胞株)共同培养,以不加任何处理因素无血清培养的肝细胞组作为对照,用MTT法及放免法观察上述各不同处理因素对正常人肝细胞的增殖的影响,统计学方法分析结果有无差异,再将上述各组细胞用流式细胞术检测细胞凋亡情况。结果1.MTT测定各处理因素中随浓度升高,对细胞增殖的抑制作用增强,其中80μg/ml的VLDL组、40μg/ml和80μg/ml的OX-VLDL及1000u/ml和2000u/ml的TNF-α组吸光度值低于对照,且统计学分析P<0.05;而且两种处理因素与肝细胞共同培养时的抑制作用比一种因素作用时增强。2.凋亡检测三种处理因素单独应用时均无明显肝细胞凋亡,但OX-VLDL与TNF-α共同作用可诱导肝细胞凋亡。结论1.肝细胞内脂质堆积及肿瘤坏死因子α过多可损伤肝细胞,两种因素共同作用于肝细胞时抑制作用增强。2.过多脂质与TNF-α共同作用可诱导肝细胞凋亡。
Objective: To explore the effects of very low density lipoprotein (VLDL), oxidized very low density lipoprotein (OX- VLDL) and tumor necrosis factor α (TNF- α) on proliferation and apoptosis of normal human hepatocyte. Methods: Normal adult hepatocytes were cultivated with VLDL, OX- VLDL and TNF- α of different concentration respectively in serum- free eultree medium. Effects of VLDL, OX - VLDL and TNF -α on proliferation of normal adult hepatocyte line were observed by means of MTF measuring respectively. Flow cytometry were used to assess apoptosis rate. Results: 1 Cell proliferation of every group was decreased.With doses increased, there was a decreased trend of cell proliferation. Cell proliferation of the VLDL group with concentration of 80μg/ml, the OX- VLDL group with 40μg/ml, 80μg/ml and TNF-α group of 1000u/ml and 2000u/ml were significant lower than control group (P 〈 0.05) .2 TNF-α, combined with OX- VLDL can induce apoptosis. Conclusion: 1 Excess of lipids and TNF - α in the liver might damage hepatocyte, and the effects of two factors together on hepatocyte were stronger than one factor alone.2.TNF- α, combined with OX- VLDL can induce apoptosis.
