摘要
从酸败果酒中分离到1株可同时利用葡萄糖和甘油产生1,3-丙二醇的乳杆菌,经鉴定为Lactobacillus brevis。当葡萄糖∶甘油为0.05mol/L∶0.1mol/L时,厌氧发酵70h,1,3-丙二醇产量为11.2mmol/L,生成速率为0.16mmol/(L.h),甘油的转化率为0.30mol/mol,甘油脱水酶活性为0.84U/mL,1,3-丙二醇氧化还原酶活性为3.80U/mL,该研究为过量生产1,3-丙二醇的乳酸菌工程菌株改造奠定了基础。
Lactobacillus sp. W5-17,isolated from spoilage wine, was identified as Lactobacillus brevis. The ratio of glucose to glycerol in the medium for 1,3-propanediol was optimized with 0. 05 mol/L : 0. 1 mol/ L. When the strain was incubated in the medium at 30℃ for 70 hours under anaerobic conditions, the concentration of 1,3-PD, the molar ratio of glycerol to 1,3-PD and the production rate were 11.2 mmol/L, 0. 30 mol/mol and 0. 16 mmol/(L· h), respectively. The maximal glycerol dehydratase activity and 1,3-propanediol NAD+ oxidoreductase activity were 0.84 U/mL and 3.80 U/mL. The strain obtained was useful for overproduction 1,3-propanediol by metabolic engineering with glycerol dehydratase and the 1,3-propanediol NAD+ oxidoreductase heteroexpression.
