摘要
目的探讨低氧诱导因子-lα(HIF-1α)在视网膜母细胞瘤(Rb)中的表达及其与血管生成的关系。方法利用免疫组织化学方法检测60例Rb、10例正常视网膜组织中HIF-1α的表达,并用CD34单克隆抗体标记血管内皮细胞,计数Rb中微血管密度(MVD)。结果HIF-1α在正常视网膜组织中未见表达,主要表达于Rb细胞浆及胞核。HIF-lα在Rb组中阳性表达率为56.7%(34/60),明显高于正常对照组(P<0.01);Rb组中MVD平均为33.59±1.15,正常对照组为7.42±0.95(t=17.499,P<0.01)。HIF-lα与MVD的表达呈显著正相关(r=0.439,P<0.01)。结论HIF-lα在视网膜母细胞瘤中的表达与肿瘤的新生血管形成有关。提示HIF-lα的表达水平可用于预测Rb的生物学行为,可作为Rb治疗的新靶点。
Objective Hypoxia inducible factor-1α (HIF-1α), a kind of transcription factor, is proved to be involved in the angiogenesis. The aim of this study was to explore the influence of HIF-1α expression on angiogenesis and the relationship between HIF-1α expression and microvessel density (MVD) with biological behavior of retinoblastoma. Methods Sixty samples of retinoblastoma tissues from surgery and 10 samples of normal retina from corpse eyes were examined for the study of HIF-1α expression by SP immunohistochemical staining,and the CD34 endothelial antigen was used to label the new microvessel. MVD in retinoblastoma tissues was calculated. Results No HIF-1α expression was found in normal retina. However,positive expression of HIF-1α was found in retinoblastoma with the positive expression rate 56.7% , showing the brown staining mainly in cytoplasm and cell nucleus (P 〈 0.01 ). The positive expression rate of HIF-1α was 39. 1% (9/23) in differentiated type of retinoblastoma and was 67.6% ( 25/37 ) in undifferentiated type of retinoblastoma( P 〈 0.05 ). The mean MVD was 33.59 ± 1.15 in retinoblastoma and 7. 42 ± 0. 95 in normal retina (t=17.499, P〈0.01). MVD was( 29.78 ±1.40),(36.91± 2.21) and (44.24 ± 1.21 ) in low intensity, moderate intensity and strong expression groups of HIF-1α, respectively, showing a positive correlation with HIF-1α expression (r = 0. 439, P 〈 0.01 ). Conclusion Angiogenesis is significantly associated with HIF-1α overexpression.
