嗜水气单胞菌产弹性蛋白酶条件的优化 被引量:2

Optimal conditions of elastase production in Aeromonas hydrophila

摘要 对嗜水气单胞菌产弹性蛋白酶条件进行如下优化:用单因素法优化了氮源、碳源等培养基成分及温度和初始pH值等培养条件;用三因素三水平的正交试验对氮源浓度、碳源浓度、盐离子浓度进行优化。结果发现:较低浓度的氮源、碳源有利于酶的产生,表面活性剂能明显提高酶的产量。嗜水气单胞菌产弹性蛋白酶的最佳条件:胰蛋白胨5 g.L-1,蔗糖5 g.L-1,NaCl 2.5 g.L-1,K2HPO42.0 g.L-1,Tween-80 1 g.L-1,初始pH为7.5,28℃培养48 h。 The conditions of elastase production in Aeromonas hydrophila strain J-1 ( Ah J-1 ) were optimized. The effects of carbon sources, nitrogen sources, temperature, initial pH, metal ion and surface activity reagent on the elastase production of Ah J-1 were investigated. The optimal condition was obtained by orthogonal experiment of three fators and three levels. The optimal medium was as follows: 5 g·L^-1 tryptose, 5 g·L^-1surose, 2.5g·L^-1 NaCl, 2.0g·L^-1 K2HPO4 Tween-80 1.0g·L^-1. In this case, Ah J-1 was cultured at 28℃ for48 h on a shaker with a speed of 100 r · min^-1.
