摘要
目的:建立家兔胆固醇结石模型,检测家兔肝组织固醇携带蛋白2(Sterol Carrier Protein,SCP2)mRNA的表达及中药对其影响。方法:将42只家兔随机分为正常对照组、高胆固醇膳食组、熊去氧胆酸组、洛伐他丁组、茵陈组和复方中药组。利用高胆固醇膳食诱发家兔胆囊胆固醇结石模型,采用半定量逆转录多聚酶链反应(RT-PCR)检测SCP2mRNA在兔肝组织的表达。结果:4周时处死动物,正常对照组无结石形成;高胆固醇膳食组7只动物有6只形成结石,成石率达85.71%;熊去氧胆酸组成石率为50.00%;洛伐他丁组成石率高达100%;茵陈组和复方中药组成石率较低,均为42.86%。各实验组与对照组相比,SCP2mRNA表达水平均有不同程度升高,各给药组动物肝组织SCP2mRNA表达差异有显著性意义。熊去氧胆酸组和洛伐他丁组SCP2mRNA表达与高胆固醇膳食组相比差异无显著性意义,茵陈组和复方中药组SCP2mRNA表达分别与高胆固醇膳食组相比差异有显著性意义(P<0.05)。除熊去氧胆酸组外,其余各组动物肝组织SCP2mRNA表达均与本组胆汁成石指数(Lithogenicindex,LI)呈正相关。结论:高胆固醇膳食致家兔胆囊结石形成时肝组织SCP2mRNA表达升高。茵陈和复方中药能通过下调SCP2mRNA的表达降低胆固醇结石的发生率;熊去氧胆酸也能降低动物的成石率,但对SCP2mRNA表达没有明显影响。
Objective:To investigate the expression of SCP2 mRNA and the effects of herbs on the expression in liver tissues of rabbit cholesterol gallstone model. Methods: Rabbit model was induced by high cholesterol diet (HCD) containing 1.2% cholesterol.42 rabbits were randomized into 6 groups:normal control group (CON group)were given normal diet, high cholesterol diet group (HCD group) were given high cholesterol diet, ursodeoxycholic group (UDC group) were given high cholesterol diet and ursodeoxycholic acid, lovastatin group (LVT group) were given high cholesterol diet and lovastatin, Yinchen group (YCH group) were given high cholesterol diet and Yinchen, and Fufangzhongyao group (FZH group) were given high cholesterol diet and Fufangzhongyao. Liver tissue was obtained from rabbits after 4 weeks and SCP2 mRNA expression was detected by using Reverse Transcription Polymerase Chain Reaction (RT-PCR). Results: After 4 weeks, CON group had no gallstone formation. 85.71% of HCD group, 50.00% of HCD group, 100.00% of LVT group,42.86% of YCH group and 42.86% of FZH group had formed stones in gallbladder. The expression level of SCP2 mRNA were all increased in 5 experimental groups as compared with CON group. There were significant difference of SCP2 mRNA expression within YCH and HCD groups, also within FZH and HCD groups. The expression level of SCP2 mRNA was respectively related to its own bile lithogenic index (LI) in CON group, HCD group,LVT group, YCH group and FZH group. Conclusions : The SCP2 mRNA of liver tissue is over-expression in HCD induced rabbit gallstone model. Yinehen can decrease the gallstone formation rate by down-regulating the expression of SCP2 mRNA.
