摘要 目的:探讨肝细胞刺激因子对骨髓单个核细胞(BMMNC)在小鼠体内向肝归巢的影响。方法:制备BALB/c小鼠2-乙酰氨基芴-四氯化碳肝损伤模型,供鼠分离BMMNC,体外经PKH26染色,经尾静脉途径输入肝损伤小鼠体内,实验组立即以80μg/(kg.d)剂量腹腔内注射肝细胞刺激因子,对照组注射等量5%葡萄糖溶液。移植2周后取小鼠肝,冷冻切片计荧光细胞数,苏木精-伊红(H-E)染色观察病理组织学变化。结果:实验组小鼠肝冷冻切片上平均荧光细胞数多于对照组(84vs61,P<0.05)。H-E切片上新生肝细胞较多。结论:肝细胞刺激因子对BMMNC向肝归巢有一定的促进作用。 Objective:To explore the effects of hepatic stimulator substance on homing to the liver of bone marrow mononuclear cells in mice. Methods:Mouse hepatic injury was induced by intraperitoned injection of carbon tetrachloride( CCl4 ) and adnunistration of 2-acetylminofluorene (2-AAF). Bone marrow stem cells seperated from donor mice were labeled with PKH26 and then were transplanted through tail vein to recipient mice whose liver were injured. HSS was injected intraperitoneal in 80μg/( kg · d) to the experiment group, and glucose was injected to the control group. Two week after transplantation, mice liver were taken out and PKH26 labeled cells were counted in frozen section . Then histological assessment was measured in H-E stained section. Results:The average of PKH26 labeled positive cells in frozen sections in HSS group were more than that in the control group ,84 vs 61 (P 〈 0.05). Regenerative hepatocyte in HSS group were more than that in the control group. Conclusion:Hepatic stimulator substance promotes homing of bone marrow mononuclear cells to the liver.
