糖尿病大鼠早期骨微结构改变及其BMP-2基因的表达 被引量:2

摘要 目的观察糖尿病大鼠早期的骨密度、骨微结构改变及骨组织中BMP-2基因的表达。方法清洁级健康成年雄性Wistar大鼠30只,按照随机数字表将其分为A组(普通饲料对照组)、B组(高脂饲料+链脲佐菌素组)和C组(普通饲料+大剂量链脲佐菌素组)。饲养12周后,分别对各组大鼠进行骨密度、光镜、扫描电镜、骨形态计量学的观察以及BMP-2基因表达等指标的检测。结果与A组相比,B组骨微结构无明显变化(P>0.05),但BMP-2基因表达增高(P<0.05),C组骨微结构明显改变(P<0.05),BMP-2基因表达降低,但差异无统计学意义(P>0.05)。结论不同的糖尿病大鼠模型早期骨微结构及BMP-2基因表达情况改变不完全一致,可能与高胰岛素血症、胰岛素抵抗有一定关系。 Objective To investigate the changes pression in rats at early period of diabetes. Methods of bone density, bone microstructure, bone BMP-2 ex- Thirty SPF 4-month-old male Wistar rats were randomly divided into 3 groups (n = 10 in each group) : A (fed with common forage as normal control group), B (fed with high-lipid forage and intraperitoneally injected with STZ to establish type 2 diabetes with insulin resistance), C (fed with common forage and intraperitoneally injected with STZ to establish type 2 diabetes with insulin secretion defaulting). The bone histomorphology, bone histomorphometry, bone BMP-2 expression were investigated 12 weeks later. Results As compared with group A, bone microstructure was of no obvious changes and bone BMP-2 expression was higher in group B ( P 〈 0.05 ) , and obvious bone breakage was observed in group C ( P 〈 0.05 ). Conclusion The changes of bone microstructure and bone BMP-2 expression at the early period of diabetes are not consistent absolutely in individual rat model of diabetes. It maybe relate to hyperinsulinism and insulin resistance.
