摘要
目的研究雌激素对大鼠局灶性脑缺血再灌注损伤后NF—ΚB蛋白和细胞凋亡变化的影响。方法采用线栓法复制局灶性脑缺血再灌注模型。应用免疫组化检测NF—ΚB蛋白表达,利用原位缺口末端标记(TUNEL)法研究凋亡细胞的变化。结果与对照组大鼠相比,雌激素组脑缺血再灌注后NF-ΚB表达明显减弱(P<0.01);凋亡细胞显著减少(P<0.01)。结论雌激素能抑制大鼠脑缺血再灌注后NF-ΚB表达,减少细胞凋亡。这可能是其脑保护作用的机制之一。
Objective To study the effects of estrogen on expression of NF-- κB and cell apoptosis in ischemialreperfusion brain tissue in ras. Methods Middle cerebra/ artery ocliusion/reper/usion rats models were performed by using modified filament method. Immunohistochemistry were used to detect expression of NF- κB. Using tunnel examines the apoptotic cells. Results Comparing with the control group, the expression of NF- κB remarkably attenuated and apoptotic cells decreased significantly in the group treated with estrogen(P〈0.01). Conclusion Estrogen inhibits the expression of NF- κB , decreases cell apoptosis after cerebral ischemia/reperfusion , this may be one of the mechanisms in which Estrogen exerts its neuro-protection.
