摘要
目的探讨氯沙坦和替米沙坦对X综合症大鼠血糖、血脂的影响。方法采用高果糖饲料饲养两肾一夹SD大鼠4周,诱导建立X综合症模型,实验随机分为正常组;两肾一夹肾性高血压组;果糖组;果糖饲养两肾一夹肾性高血压组;氯沙坦组和替米沙坦组。每周检测各组尾动脉收缩压,于12周末采血测定各组血糖、血脂、血胰岛素水平,同时计算胰岛素敏感指数。结果氯沙坦组与果糖饲养两肾一夹肾性高血压组对比,氯沙坦降低了收缩压(P<0.01)、空腹血糖(8.01 mmol/L比13.81 mmol/L,P<0.01)、甘油三酯(0.88 mmol/L比1.53 mmol/L,P<0.01)、总胆固醇(0.79 mmol/L比1.57 mmol/L,P<0.01),升高了胰岛素敏感指数(-5.71比-6.40,P<0.05);而替米沙坦组与果糖饲养两肾一夹肾性高血压组对比,替米沙坦降低了收缩压(P<0.01)、空腹血糖(8.52 mmol/L比13.81 mmol/L,P<0.01)、血胰岛素(28.66 MIu/L比37.69 MIu/LP<0.05)、甘油三酯(0.63 mmol/L比1.53 mmol/L,P<0.01)、总胆固醇(0.99 mmol/L比1.57 mmol/L,P<0.01),升高了胰岛素敏感指数(-5.50比-6.40,P<0.05)、高密度脂蛋白(1.03 mmol/L比0.78 mmol/L,P<0.05)。结论氯沙坦和替米沙坦能降低X综合症大鼠的血压,减轻胰岛素抵抗,纠正糖、脂代谢紊乱。
Objective To investigate the effect of losartan and telmisartan on blood sugar and blood lipid in syndrome X rats. Methods Syndrome X was produced by two - kindey, one - cliped(2K1C)rats fed with high fructose diet for 4 weeks. The rats were randomly divided into six groups: Control, 2K1C - RHR, HFD, 2K1C - RHR + HFD, Losartan and Telmisartan. Systolic blood pressure(SBP) was measured each week, fasting blood sugar (FBS), blood lipid, and the level of fast serum insulin(FSI) were measured respectively in groups at the time of 12 weekend, and insulin sensitive index(ISI)was also calculated. Results Comparing losartan group with 2K1C - RHR+ HFD group, losartan decreased systolic blood pressure( P 〈 0. 01), fasting blood Sugar (8. 01 mnol/L vs 13.81 lnmol/L P 〈 0.01 ), triglycelide (0.88 mmol/L vs 1.53 mmol/L, P 〈0.01 ), blood total cholesterol ( 0. 79mmol/L vs 1.57 mmol/L, P 〈 0.01 ), raised insulin sensitive index ( - 5.71 vs - 6.40, P 〈 0.05 ) ; Then, compar- ing telmisartan group with 2K1C - RHR + HFD group, telmisartan also decreased systolic blood pressure( P 〈 0.01 ), fasting blood Sugar ( 8.52 mmol/L vs 13.81 mmol/L, P 〈 0.01 ), fasting serum insulin( 28.66 MIu/L vs 37. 69 MIu/L, P 〈 0.05 ), triglyceride( 0.63 mmol/L vs 1.53 mmol/L, P 〈 0.01 ), blood total cholesterol (0.99 mmol/ L vs 1.57 mmol/L, P 〈 0.01 ), increased insulin sensitive index(- 5.50 vs - 6.40, P 〈 0.05 ), high density lipoprotein cholesterol( 1.03 mmol/L vs 0.78 mmol/L, P 〈 0.05). Conclusions Losartan and Telmisartan can reduce BP, improve insulin resistance and correct the abnormity of sugar and lipid metabolism.
