摘要
分析了云南大叶茶种和福鼎白毛茶及其F1代的42品系的茶氨酸含量,挑选出茶氨酸含量高的5个材料与茶氨酸含量低的5个材料,SDS法提取茶树基因组DNA,用140个随机引物对这二组材料进行RAPD扩增,其中14个引物在高茶氨酸材料与低茶氨酸材料间可扩增出多态性产物,并寻找到与茶树高茶氨酸含量相连锁的RAPD标记OPB20-2784bp,为茶树分子标记辅助育种和早期鉴定提供了方便快捷的检测技术。
Based on the theanine analysis of tea hybrid F1 42 cultivars originated by Yunkan 10# ( C. assamica) and Fuding ( C. sinensis), 5 higher theanine and 5 lower theanine cultivars were supplied for RAPD-PCR. And then DNA of hybrid F1 10 cultivars were isolated by SDS ( Sodium Dodecyl Sulphate ) method,14 RAPD primers were polymorphic between higher and lower theanine group from 140 RAPD primers in RAPD-PCR, among them primer OPB-20 linked to higher content theanine gene was discovered during many times repetition.
