摘要
目的:探讨补肾活血方对恒河猴PCO模型卵巢局部纤溶酶原激活物抑制因子-1(PAI-1mRNA)表达的影响。方法:选择健康育龄雌性恒河猴随机分为正常组、模型组、中药组;采用皮下注射丙酸睾丸酮和绒毛膜促性腺激素方法建立PCO模型;分别灌胃给予生理盐水、补肾活血方,采用原位杂交技术观察其卵巢局部PAI-1mRNA表达的变化。结果:恒河猴卵巢PAI-1mRNA的表达主要存在于卵泡膜细胞及间质细胞,模型组表达强度超过正常组,中药组表达水平明显低于模型组,与正常组相比无差异(P>0.05)。结论:恒河猴PCO模型卵巢局部PAI-1mRNA异常表达,补肾活血方具有改善这种异常表达作用,可能是其促进卵巢排卵的作用机制。
Objective:To observe the change of ovarian PAI-1mRNA in monkey with polycystic ovary(PCO)and approach the mechanism of bushen houxue prescription on it.Methods:PCO model was built by injecting testosterone propionate and HCG.normal,model and treatment groups were given saline and bushen huoxue prescription respectively.Ovarian PAI-1mRNA expression change was determined by situ hybridization technique. Results:The expression of ovarian PAI-1mRNA was more obvious than that of normal group,the expression in the treatment group was in- hibited,there was no difference between the treatment group and the normal(P>0.05).Conclusion:the expression of ovarian PAI-1m- RNA in PCO monkey was abnormal,bushen huoxue prescription can improve this abnormal expression.
