奶牛血液保存条件和DNA提取方法的优化 被引量:4

摘要 本文研究中国荷斯坦牛血液在不同温度、时间保存的条件下(4℃保存0d为A组;-70℃保存30d为B组;4℃保存7d为C组)对DNA提取效果的影响和对比试剂盒法(E)、试剂盒改良法(F)、酚-氯仿法(G)提取中国荷斯坦牛血液基因组DNA的效果。试验结果表明:A组与B组差异不显著(P>0.05);A组、B组与C组差异显著(P<0.05)。4℃保存0d对DNA提取效果影响最小。在A组中,E与F差异不显著(P>0.05);E、F与G差异显著(P<0.05)。综合各方面因素,试剂盒改良法最好。 This article study on effects of different temperatures and storage time (group A:4℃ 0 day; group B:-70℃ 30 days; group C:4℃ 7 days) on blood DNA extracting from Chinese Holstein and comparison of three blood DNA extraction methods [reagent box method(E); improved reagent box method (F);phenol-chloroform method (G)].The results indicate:it had no significant effect on blood DNA extraction between group A and group B (P〉0.05);it bad significant effect on blood DNA extraction between group A and group C (P〈0.05),also between group B and group C. when the blood was group A,It has the least influence on blood DNA extracting. In group A, it had no significant effect on blood DNA extraction between using method E and method F (P〉0.05); It had significant effect on blood DNA extracting between using method E and method G(P〈:0.05),also between using method F and method G.All factors considered, improved reagent box method is the best method of the three methods.
