摘要 为了研究DM-2型大孔树脂精制苦荞总黄酮的优化工艺条件,以树脂对苦荞总黄酮的吸附率、解吸率为评价指标,采用响应曲面法(response surface methodology,RSM)建立树脂法精制苦荞总黄酮的二次多项回归模型方程,对精制苦荞总黄酮吸附解吸工艺条件进行优化。结果表明吸附最佳工艺条件为吸附速率2 mL/min,pH值2,上样液浓度2 mg/mL。解吸最佳工艺条件:乙醇浓度71.05%,pH值8.76,解吸速率1 mL/min。该方法简单可行,精制效果好,适合于工业化生产。 In order to study the purifying technology of tartary buckwheat total flavonoids with DM-2 macroporous resin, the adsorption rates, desorption rates of macroporous resin were selected as monitoring parameters for separating efficiency. Response surface methodology(RSM) was employed in the study and a mathematical regression model for purification of tartary buckwheat total flavonoids with the macroporous resin was built. The technological parameters of the purifying process of tartary buckwheat total flavonoids with the macroporous resin were optimized. Results show that the optimal absorption parameters are adsorption velocity 2.0 mL/min, pH 2, solution concentration 2 mg/mL and the optimal desorbtion parameters are 71.05% ethanol as eluting solvent, pH 8. 76, the desorption velocity I mL/min. This method is simple and feasible with good effect of purification, which can meet industrial requirements.
