摘要
焦磷酸测序(pyrosequencing)技术是一种新的DNA序列分析技术,其特点是操作简便、大通量、自动化,适合大量样本的快速检测,无须进行电泳、DNA序列无须荧光标记等,是一个理想的遗传分析技术平台。文章对焦磷酸测序技术原理及近年来在单核苷酸多态性研究、微生物鉴定分型和DNA甲基化等方面的应用作一简要综述。
Pyrosequencing is a DNA sequencing technique based on the detection of released pyrophosphate (PPi) during DNA synthesis. Pyrosequencing is characterized by easy handling, high throughput and automation. This article discusses the principles and the operating procedures of this new technology and the latest advances in its application in single nucleotide polymorphism (SNP) genotyping, microbial typing and CpG methylation analysis.
