摘要
给雏鸡一次投服亚硒酸钠0.6%mg/kg·BW后,不同时间采集血样,用荧光分光光度计测定血硒含量。在IBM微机上用药国学程序软件(MCPKP)处理血硒含量实测值,获得血硒浓度理论值及药动学参数。血清经时过程符合一级吸收二室开放模型,理论方程为:Ci=0.1160e-0.1476t+0.0119e-0.200t-0.1279e-0.8628t。主要动力学参数(均值):吸收相半衷期消除半衰期,t1/2kα=0.80(h);达峰时间Tmax=2.458(h);达峰浓度Cmax=0.0768(μg/ml)分布相半衰期t12α=4.69(h);消除半衰期,t12β=34.63(h).并根据单剂量给药参数计算出多剂量给药参数(叠合法公式),提出对雏鸡用药方案,先导剂量为1.5739mg/kg·BW,维持剂量为0.6mg/kg·BW,给药间隔时间τ=24(h),血药平均稳定浓度(c)为0.0513μg/ml。
ollowing singal oral dose of sodium selenite solution(0.6mg/kg·BW),the plood samles were collectdt at different intervals.Theselenium concentration in blood were measured by Fluori-spestrophotoneter.The rtue values of selenium concentration wereprocessed with MCPKP wich is special designed for the pharmacokinetieson compuler 1BM,and by which the theoretical values of seleniumconcentration in blood and the pharmacokinetics parameters wereobtained.the lood selenium concentration-tine curve could be deseribedby the two-compartment open modle with first order absorption.The drug-time equation is Ci=0.1160e -0.1476t +0.0119e -0.0200t +0.1279 -8628t The main kineties parameters(average valuc)are as following thehalfllife of obsorption(t1/2 k α)is 0.80 h,the tine of reachingmaxinum concentration(tmax)is 2.58 h,the maxiinum concentration(Cmax)is 0.076 ug.ml,the half-life of distribution(t1/2α)is 4.69 h.the half-life of elimination(t1/2α)is 34.63 h.Based on singaldosing parameters,multiple dosing parameters were caculted(Multipledosing parameler formula)and drug schedule give to the chicken wasconducted.the first dosing is 1.5739 mg/kg·BW.themaintenane dosing is 0.6 mg/kg·BW,the interval tine of drug(γ)is 24 h, the average steady-state plsdma concentration(C)is 0.05ug/ml.
