植物生长调节剂和AgNO_3对大白菜外植体不定芽再生的影响

摘要 通过L25(56)正交试验,优化及筛选最适合大白菜'鲁白6号'子叶及下胚轴不定芽诱导的4种植物生长调节剂和AgNO3的浓度组合。结果发现,在α-NAA、6-BA、TDZ、ABA和AgNO3中,α-NAA对子叶不定芽诱导率的影响达到5%水平显著差异,确定以MS+1mg/Lα-NAA+2mg/L TDZ+5mg/LAgNO3为最佳子叶不定芽诱导培养基,MS+0.8mg/Lα-NAA+5mg/L6-BA+1mg/L TDZ+7mg/L AgNO3为最佳下胚轴不定芽诱导培养基。 The cotyledon and hypocotyl of Brassica campestris ssp.pekinensis Lubai'6 were cultured for adventitious-bud induction on MS basic medium containing different concentrations of α-NAA,6-BA,TDZ,ABA and AgNO3 with orthogonal design L25(56).The results indicated that α-NAA was important to the adventitious-bud induction of cotyledon.The optimal adventitious-bud induction medium of cotyledon was MS medium with 1 mg/L α-NAA,2 mg/L TDZ and 5 mg/L AgNO3,and the optimal adventitious-bud induction medium of hypocotyl was MS medium with 0.8 mg/L α-NAA,5 mg/L 6-BA,1 mg/L TDZ and 7 mg/L AgNO3.
