摘要
针对金黄色葡萄球菌与氟喹诺酮类药物耐药相关的外输泵基因norA,构建了含有norA基因的pUT-norA多重耐药载体及相应的金黄色葡萄球菌多重耐药菌株,以所构建菌株为试验菌株,应用浸渍法、煎煮法制备了64种中药提取物,经联合药敏试验,从无抑菌作用的50种中药提取物中筛选出对金黄色葡萄球菌耐药性有抑制作用的外输泵蛋白抑制剂:浙贝母、射干、穿心莲和菱角。
Multi-drug efflux pump is an important mechanism in multi-drug resistance Staphylococcus aureus. The norA gene was cloned into the expression vector, which is co of vector pUB-110 and pMD-18T. Quinolone-susceptible S. aureus was transformed with the resulting plasmid, pUTnorA. In conclusion, we have constructed multi-drug resistance S. aureus, in which genetics and mechanisms of quinolone resistance was known. 64 kinds of apozems were extracted from Chinese herbs by pickling process and decocting method. The bacteriostasis effectiveness was observed by punch method and broth dilution method in 64 kinds of Chinese herbal extraction. Bacteriostatic results of medicine and CPFX indicated that Fritillaria thunbergii, Belamcanda chinenois, Andrographis paniculata and Trapamanshurica tier contain inhibitor of efflux.
