摘要
目的:应用Walker-256细胞建立大鼠骨癌痛模型。方法:将4×104个W256癌细胞注入SD大鼠胫骨上段骨髓腔内,利用大鼠热板法和足跖加压法分别测量热刺激和机械刺激痛阈的变化,X线摄片进行放射学评估骨质破坏程度,同时,组织切片HE染色观察肿瘤生长和骨结构的破坏情况。结果:造模动物在12天左右开始出现热刺激和机械刺激痛阈明显下降(痛觉过敏),并呈渐进性发展;胫骨X线摄片显示14天即有明显的骨破坏,第21天时出现病理性骨折;组织切片显示骨髓腔内肿瘤生长活跃,向外侵蚀破坏骨皮质。结论:从疼痛行为学、放射学、组织学等方面的研究结果显示,应用Walker-256细胞成功建立了大鼠骨癌痛模型。
Objective : It is to establish a useful animal model of bone cancer pain by using Walker - 256 tumor cells. Methods : A rat model of bone cancer pain was produced by intra - tibial inoculation of 4×10^4 Walker - 256 tumor cells in Sprague - Dawley rats. Pain threshold of thermal stimulation was measured by hot plate tester,and pain threshold of mechanical stimulation was measured by paw - pressure detector. The development of the bone tumor and structural damage to the bone was examined with radiological and histology analysis. Results : Rats with intra - tibial injections of Walker - 256 tumor cells displayed the gradual development of decrease in the pain threshold from thermal and mechanical stimulations because of hyperalgesia, which began on the 12^th day after injection of the tumor cells. The radiograph of the tibial showed that clear bone destruction happened on the 14^th day and pathological fracture happened on the 21^st day. Sections of the bone showed the bone marrow cavity was full of tumor cells, which were proliferating actively and destructing the cortical bone. Conclusion: Studies in terms of ethology, radiology and histology have showed that this model of bone cancer pain has been established successfully by using Walker- 256 tumor cells.
