凉血散瘀汤对实验性脑出血大鼠的ET TNF-α FIB和TT影响 被引量:2

摘要 目的:探讨凉血散瘀汤治疗实验性脑出血大鼠的可能机制。方法:将60只Wistar大鼠以随机表法随机均分6组:空白组、假手术组、模型组、治疗1组,治疗2组、治疗3组。后4组冬采用2次注射自体动脉血法构建大鼠脑出血模型,术后3h治疗1、2和3组分别给予凉血散瘀汤2.4、4.8和7.2mg/(kg.d)灌胃,其他3组给予等量生理盐水。应用放射免疫法测定血ETand TNF-α含量。结果:与空白组比较,假手术组血ET和TNF-α轻度升高(P>0.05);与假手术组比较,模型组血ET和TNF-α明显升高(P<0.01);与模型组比较,治疗组血ET和TNF-α明显下降(P<0.01);治疗组间比较血ET和TNF-α下降差不多。模型组和治疗组FIB水平较正常组升高(P<0.01)。模型组和治疗组TT与正常组比较并无明显差异(P>0.05)。结论:凉血散瘀汤治疗大鼠脑出血后对神经功能具有保护作用,其机制可能抑制ET表达、TNF-α毒性和降低FIB活性有关。 Objective :To study possible mechanism of Liangxuehuayu method in the treatment of the rats of intmcerebral hemorrhage. Methods :60 Wistar rats were randomized into six groups:normal group, sham group, model group, treatment group one treatment group two and treatment group three. The rats in the latter four groups were subjected to intracerebral hemorrhage models by injecting autologous arterial blood into caudate nucleus-with double - injection method. Three hours after modeling,treatment group one were given"Liangxueshangyutang" at the dosage of 2.4mg/( kg·d) ,treatment group two given 4.8mg/( kg·d), and treatment group three given 7.2mg/( kg·d). Treatment groups were given consecutive 3davs. The other 3 groups were given normal saline at the dosage of 2.4mg/( kg·d). The contents of blood ET and TNF-α were detected by radioimmunoassay. Results: In comparison with normal group,the contents of ET and TNF-α in sham group rose lightly( P 〉0.05) ;in comparison with sham group,the contents of ET and TNF -α in model group rose remarkably( P 〈0.01 ) ;in comparison with model group,the contents of ET and TNF - α in trealment groups fell remarkably( P 〈 0.01 ) ; among the treatment groups, the contents of ET and TNF - α was almost equally fallen. The level of FIB in model and treatment groups is higher than in normal group( P 〈 0.01 ). Howeverthe level of TT in model and treatment groups is not significantly different from the normal group( P 〉0.01 ). Conclusion: Liangxueshangyutang in the treatment after intracerebral hemorrhage can protect nerve functions possibly by suppressing ET and TNF - α and decreasing the activation of FIB.
