卡介菌多糖核酸对肺间质纤维化大鼠超微结构的影响 被引量:2

摘要 目的:观察卡介菌多糖核酸对肺间质纤维化大鼠肺组织超微结构的影响。方法:实验于2006-03/06在锦州医学院药理实验室完成。36只大鼠随机分为3组,每组12只:①正常对照组:不干预。②模型组:气管内一次性注入盐酸平阳霉素5mg/kg,制成肺间质纤维化模型。③卡介菌多糖核酸组:同模型组造模,于造模第2天开始,腹腔注射卡介菌多糖核酸0.5mg/只(1g/L),隔日1次。以上各组于实验第7,28天各处死6只,进行支气管肺泡灌洗并收集灌洗液。在透射电镜下观察各组大鼠肺组织超微结构的变化,并通过测定肺泡灌洗液中细胞计数和肺组织中羟脯氨酸水平来评价干预效果。结果:36只大鼠进入结果分析。①超微结构的变化:模型组28d时Ⅱ型细胞数目较前减少,其内板层小体减少,呈空泡样变,纤维母细胞增生,胶原纤维明显增多,卡介菌多糖核酸组较模型组明显减轻。②羟脯氨酸水平:卡介菌多糖核酸组7,28d时均低于模型组[7d:(1.44±0.08),(1.61±0.17)μg/L;28d:(1.74±0.04),(2.32±0.07)μg/L;P<0.05],但仍高于正常对照组(P<0.01)。③肺泡灌洗液中细胞总数模型组各期均高于正常对照组(P<0.01),卡介菌多糖核酸组低于模型组(P<0.01),两组均显示第7天细胞总数最高;模型组及卡介菌多糖核酸组中性粒细胞及淋巴细胞比例均高于正常对照组,但卡介菌多糖核酸组较模型组减少。结论:卡介菌多糖核酸能减轻大鼠肺间质纤维化。 AIM: To study the influence of Bacillus Calmette Guerin (BCG-PSN) on the ultrastructure of pulmonary tissue in rats with pulmonary fibrosis. METHODS: The experiment was conducted in the Laboratory of Pharmacology, Jinzhou Medical College between March and June 2006. Thirty-six rats were randomly divided into 3 groups with 12 rats in each group: (1) Normal control group: Animals received no intervention. (2) Model group: Rats were made into models of pulmonary fibrosis by intratracheal injection of bleomycinA5 (5 mg/kg).(3) BCG-PSN group: Rats were established into models the same as the model group, and abdominally injected with BCG-PSN since the second day of modeling once every other day, and 0.5 mg for each rat. Six rats in each group were executed on the 7^th and 28^th days to collect the bronchoalveolar lavage, and the changes in ultrastructure of pulmonary tissues were observed under the transmission electron microscope in each group. The effect was evaluated by testing the cell numbers and the level of hydroxyproline in pulmonary tissues. RESULTS: A total of 36 rats were involved in the analysis of results. (1) Changes in ultrastructure: The numbers of type Ⅱ cells in the model group on the 28^th day decreased than those before, and inner lamellar bodies decreased with vacuole-like changes; fibroblast proliferated and the collagen fibers significantly increased, which was remarkably less in the BCG-PSN group than those in the model group. (2) Level of hydroxyproline: It was lower in the BCG-PSN group on the 7^th and 28^th day than that in the model group [7^th day: (1.44±0.08), (1.61±0.17) μg/L; 28^th day: (1.74±0.04), (2.32± 0.07) μg/L; P 〈 0.05]. However, it was still higher than that in the normal control group (P 〈 0.01 ), while it was lower in BCG-PSN group than that in the model group (P 〈 0.01), and it was the highest on the 7th day in both groups. The proportion of neutrophilic leukocyte and leukomonocyte in the model group and BCG-PSN group were higher than that in the normal control group, but it was less in BCG-PSN group than that in the model group. CONCLUSION: BCG-PSN can obviously alleviate pulmonary fibrosis in rats.
