硫辛酸对D-半乳糖致过氧化损伤小鼠海马细胞DNA的影响 被引量:1

摘要 [目的]观察摄入不同剂量硫辛酸对D-半乳糖致过氧化损伤小鼠海马细胞DNA的影响。[方法]清洁级雌性昆明种小鼠按体重随机分为5组,除空白对照组外,其余4组用D-半乳糖颈背部皮下注射,1次/d,连续6周。其中1组为模型组,余3组为硫辛酸干预组;其后经口给予不同浓度硫辛酸,空白组、模型组给予同体积溶剂,同时,模型组和干预组继续给予相同剂量的D-半乳糖颈背部皮下注射,连续8周。干预结束后,用单细胞凝胶电泳法检测和评价各组小鼠海马细胞DNA损伤。[结果]模型组小鼠海马彗星细胞尾长、尾矩、Olive尾矩值均最大,对照组均最小(P<0.05),干预组各指标均小于模型组(P<0.05)。[结论]硫辛酸是一种有效的抗氧化剂,对小鼠海马细胞DNA损伤有修复作用。 [Objective] To observe effects of lipoic acid on DNA oxidative damage of mice hippocampus caused by Dgalactose. [Methods] Pure Kunming mice were divided into 5 groups (1 blank, 1 model group and 3 experiment groups) randomly. Experiment groups and model group took D-galactose through subcutaneous injection once per day for 14 weeks. In the last 8 weeks, the 3 experiment groups were given different doses of lipoic acid by oral administration. DNA oxidative damage of mice hippocampus of all groups was evaluated by using single cell gel electrophoresis. [Results] Tail-length,Tail-moment and olive-tail moment of comet-like cells in hippocampus of model group were longer than those of other groups (P〈0.05) and All indexes of 3 experiment groups were shorter than those of model group. [Conclusion] Lipoic acid is an effective antioxidant,it can protect mice hippocampus DNA from oxidative damage.
