羽毛粉酶法水解工艺条件优化的研究 被引量:16

摘要 采用蛋白酶水解羽毛粉对蛋白酶进行筛选,并对酶解的工艺条件进行优化研究,分析了酶的种类、酶的添加量、缓冲溶液、温度、pH、反应时间等因素对羽毛粉水解的影响。结果表明:对酶解羽毛粉最适作用的酶是木瓜蛋白酶,木瓜蛋白酶最适反应缓冲溶液是柠檬酸-柠檬酸钠缓冲溶液,在温度为60℃,pH 5.6,作用时间8h,羽毛粉∶酶干粉=100∶2时,可得到较好的效果。此时,水解上清液氨基氮含量为24.932 mg/L,酶化效果为107.8%。 The proteinase type was selected by analyzing effects of proteinase type, proteinase added amount, buffer solution, temperature, pH, reaction time and other factors on hydrolysis of feather meal to optimize the technological conditions for proteinase hydrolysis. The results showed that the papain was the suitable enzyme for hydrolysis of feather meal and the amino nitrogen content of upper hydrolysis solution and enzyme reaction effect were 24. 932 mg/L and 107.8% under the conditions of buffer solution (C6H8O7·H2O-Na3C6H5O7·2H2O), at 60℃, pH 5.6, eight hours, and ratio of feather : enzyme=100 : 2.
