摘要 目的研究磷脂酰肌醇-3激酶(PI-3K)在H2O2诱导PC12细胞凋亡中的作用。方法采用H2O2诱导PC12细胞制作凋亡模型,通过MTT法、Hoechst/PI荧光双染及流式细胞术分析使用PI-3K特异抑制剂LY294002预处理对H2O2诱导PC12细胞凋亡作用的影响。结果100μMH2O2处理PC12细胞24 h,细胞出现明显凋亡;LY294002预处理1 h后凋亡细胞数增多(P<0.05)。结论PI-3K对H2O2诱导的PC12细胞凋亡具有抑制作用。 Objective To investigate the effect of phosphatidylinositol 3 kinase (PI -3k) signal pathway on apoptosis induced by H2O2 in rat pheoehromoeytoma(PC12) cells. Methods Pretreated with or without LY294002 which is the specific inhibitor against PI- 3k, PC12 cells were used as the H2O2 - induced apoptosis model in vitro by MTT, fluorescence microscopy using Hoeehst33342/PI nuclei staining and flow eytometry. Results Apoptosis took place after PC12 cells had been treated with 100μM H2O2 for 24hr. LY294002, a specific inhibitor of PI - 3K, enhanced H2O2 -indueed PC12 cell apoptosis. Conclusion PI -3K plays a crucial role in prohibiting PC12 cell apoptosis induced by H2O2 .
