摘要
目的:研究红景天苷对PC12细胞凋亡的影响,并探讨其抗凋亡的机制。方法:以NaCN及缺糖诱导的PC12细胞凋亡为模型,采用MTT比色法,琼脂糖凝胶电泳分析及流式细胞技术,考察红景天苷对细胞凋亡的保护作用;采用比色法测定Caspase活性、Western-blot技术检测胞浆中细胞色素C水平,考察红景天苷抗凋亡的机制。结果:红景天苷可以有效地改善NaCN及缺糖诱导的PC12细胞凋亡,抑制Caspase活性,降低胞浆中细胞色素C的水平。结论:红景天苷可能通过线粒体途径抑制NaCN及缺糖诱导的PC12细胞凋亡。
Aim: To investigate the protective effects of salidroside on injured nerve cells by an in vitro model of neuronal impairment treating PC12 cells with NaCN and glucose deprivation and its mechanism of anti-apoptosis. Methods: NaCN and glucose deprivation-induced PC12 cells apoptosis model was established, and the protective effect of salidroside were assayed by MTT, agamse gel electmphoresis and flow cytometry. Furthermore, the anti-apoptosis mechanism of salidroside was investigated. Caspase activities were measured colorimetrically and the level of cytochrome C was detected using Western-blot. Results: Salidroside has effective function in protecting impaired nerve cells and inhibiting apoptosis, also Caspase activity was repressed and level of cytochrome C was decreased. Conclusion: Salidroside may play an important part in protecting nerve cells from apoptosis induced by NaCN and glucose deprivation by mitochondria pathway.
