应用大孔吸附树脂纯化川芎有效部位 被引量:12

摘要 目的探讨大孔吸附树脂对川芎提取物的纯化条件及纯化效果。方法以阿魏酸作为考察指标,通过阿魏酸在树脂上的吸附量和解吸率筛选树脂的种类;以阿魏酸的转移率为指标考察药液的pH值、上柱吸附流速、树脂药材质量比、树脂柱径高比、清洗液的pH值、清洗液用量及流速、洗脱液种类、洗脱液用量及流速等纯化条件对纯化川芎效果的影响,与醇提水沉法进行纯化效果的比较。结果采用HPD300大孔吸附树脂作为吸附树脂,树脂药材质量比为2∶1,树脂柱径高比为1∶7,上柱药液和清洗液pH值为3.0,质量分数为50%的乙醇溶液作为洗脱液,上柱吸附流速为每小时2倍床体积,清洗和洗脱流速为每小时8倍床体积。在该纯化条件下,阿魏酸的转移率为91.67%,与醇提水沉法相当;固形物中阿魏酸的含量约为醇提水沉法的4.1倍,固形物的质量(固形物得率)比醇提水沉法减少了约76%。结论通过纯化条件的优化,大孔吸附树脂法可提高阿魏酸的纯化效果,优于醇提水沉法。 Objective To study the conditions and effects of purification of the extract of Ligusticum chuanxiong Hort. by adopting macroporos resin. Methods The kinds of resin were selected through the determination of adsorption ratio and elution ratio of ferulic acid in resin. The reservation ratio and transfer ratio of ferulic acid were used as index to investigate the purification conditions such as the pH value of water extract, the flow speed for adsorption, the ratio of the resin to material drug, the ratio of diameter to height of resin column, the pH value and flow speed of cleaning agent, the kinds and flow speed of the elution agent, etc. The results of purification of macroporous resin were compared with the water sedimentation. Results HPD300 was selected as the adsorbed resin, the ratio of resin to material drug was 2 : 1, the ratio of diameter to height of resin column was 1:7, the pH value of water extract and cleaning agent were adjusted to 3.0, 50% alcohol was chose as elution agent, the flow speed of adsorption was twofold of bed volume per hour, the flow speed of cleaning and elution was eightfold of bed volume per hour. Under the purification condition, the transfer ratio of ferulic acid was 91.67 %, which was comparable to that of the alcohol sedimentation method. Compared with the water sedimentation method, the macroporous resin purification method could increase the content of ferulic acid in the dried solid about 4.1 times, while reducing the weight of dried solid about 76 %. Conclusions The method of macroporous resin purification is superior to the method of water sedimentation.
